General description
Inhibitor of growth protein 4 (UniProt Q9UNL4; also known as Brain my036 protein, Candidate tumor suppressor p33 ING1 homolog, p29ING4) is encoded by the ING4 gene (ORF My036; Gene ID 51147) in human. The inhibitor of growth (ING) family tumor suppressor ING4 is a transcription regulator that exerts its function via chromatin remodeling. ING4 to targets the trimethylated lysine 4 of histone H3 (H3K4me3) via its conserved plant homeodomain (PHD) finger motif and is shown to associate with the HBO1/JADE histone acetyltransferase (HAT) complex. ING4 is shown to play a role in many cancer-related cellular processes, including cell proliferation, apoptosis, migration, angiogenesis, contact inhibition, DNA damage response, and hypoxia. Both gene deletion and reduced ING4 expression are reported in cancers of diverse tissue types, although ING4 knockout by itself does not cause increased spontaneous tumor formation in mice. Immunohistochemistry analysis of breast cancer tissues shows that 34% of tumors expressed undetectable to low levels of ING4. In addition, ING4 downregulation inversely correlates with an increase in NF-κB activation and NF-κB-dependent genes expression in primary breast tumors.

Specificity
Clone BTIM-4 reacts with human and murine ING4, but not human ING1, ING2, or ING5 (Byron, S.A., et al. (2012). PLoS One. 7(10):e46823). Expected to react with human ING4 isoform 1 (ING4_v1) and isoform 8 (deltaEx6A). Reactivity toward human ING4 spliced isoform 2 to 7 has not been tested.
