General description
Mitogen-activated protein kinase 14 (EC 2.7.11.24; UniProt P47811; also known as CRK1, MAP kinase 14, MAP kinase p38 alpha, MAPK 14, MAX-interacting protein 2, Mitogen-activated protein kinase p38 alpha, p38a, p38alpha, p38MAPK, SAPK2a, Stress-activated protein kinase 2a) is encoded by the Mapk14 (also known as Crk1, Csbp1, Csbp2, Mxi2) gene (Gene ID 26416) in murine species. p38 mitogen-activated protein kinases (MAPKs) constitute a group of stress-activated serine/threonine-specific kinases (SAPKs), including p38α (MAPK14/SAPK2A), p38β (MAPK11/SAPK2B), p38γ (ERK6/MAPK12/SAPK3), and p38δ (MAPK13/SAPK4), that mediate cellular signal transduction and play a vital role in numerous biological processes. p38α and p38β are ubiquitously expressed, while p38γ and p38δ are differentially expressed depending on tissue type. Sequence comparisons show approximately 60% identity within the p38 group. but only 40-45% to the other three MAP kinase family groups, extracellular signal-regulated kinases (ERKs), c-jun N-terminal or stress-activated protein kinases (JNK/SAPK), and ERK/big MAP kinase 1 (BMK1). All four p38 kinases contain a conserved Thr-Gly-Tyr (TGY) dual phosphorylation motif and are activated by dual kinases called MAP kinase kinases (MKKs).
